醫(yī)學(xué)論文范文:Toll樣受體2對角質(zhì)形成細(xì)胞分泌IL8的影響
【摘要】 目的:觀察白念珠菌刺激人角質(zhì)形成細(xì)胞后IL8的變化以及Toll樣受體2(Tolllike receptor 2,TLR2)對IL8分泌的影響,探討角質(zhì)形成細(xì)胞在皮膚免疫系統(tǒng)中的作用。方法:白念珠菌分別與抗TLR2抗體預(yù)處理前后的角質(zhì)形成細(xì)胞孵育,ELISA方法檢測不同時(shí)間培養(yǎng)上清液IL8的水平。結(jié)果:白念珠菌可引起IL8明顯升高(P<0.01),3h開始,至24h達(dá)到高峰;中和TLR2受體后,雖然引起IL8升高,但無統(tǒng)計(jì)學(xué)意義(P>0.05)。結(jié)論:白念珠菌可以刺激角質(zhì)形成細(xì)胞分泌IL8,TLR2在角質(zhì)形成細(xì)胞IL8表達(dá)中的作用尚難確定。

Abstract: Objective To observe the change of Interleukin8(IL8) in keratinocytes induced with Candida Albicans and the effect of Tolllike receptor2(TLR2) on IL8 secretion,to explore the role of keratinocytes in skin immune system. Methods Candida Albicans were cultured on keratinocytes treated before and after antiTLR2 monoantibody(mAb),respectively.The levels of IL8 in culture supernatant were determined with ELISA at different time points. Results The IL8 levels were significantly increased after induced with Candida Albicans(P<0.01),the significant increase was observed at 3 h and reached the peak at 24 h;the increase of IL8 levels had no significant difference after TLR2 was neutralized with antiTLR2 mAb(P>0.05). 念珠菌在人體表面的分布廣泛而常見,而白念珠菌是引起念珠菌病的主要致病菌種。在去除表皮的實(shí)驗(yàn)說明表皮角質(zhì)形成細(xì)胞在抵抗念珠菌感染中起重要作用。皮膚粘膜念珠菌病常表現(xiàn)為病原菌寄生部位的角層下膿皰或角層內(nèi)大量中性粒細(xì)胞聚集,但是否與細(xì)胞表達(dá)的趨化因子IL8有關(guān),目前還不太清楚[1]。Toll樣受體2(Tolllike receptor 2,TLR2)是表達(dá)于角質(zhì)形成細(xì)胞并參與真菌產(chǎn)物識別的天然受體蛋白,其與IL8的關(guān)系亦研究不多,故本研究旨在探討TLR2對角質(zhì)形成細(xì)胞分泌IL8的影響。
