醫(yī)學(xué)論文范文:環(huán)氧化酶-2及其選擇性抑制劑塞來昔布與舌鱗狀細胞癌侵襲的研究進展
【摘要】 環(huán)氧化酶(COX)是致炎物質(zhì)地諾前列酮合成過程中的重要限速酶。COX-2是環(huán)氧化酶的誘導(dǎo)型,參與炎癥反應(yīng)和腫瘤的發(fā)生。包括舌鱗狀細胞癌在內(nèi)的大多數(shù)惡性腫瘤均有COX-2的陽性表達。COX-2的活性增強還能提高細胞與細胞外基質(zhì)蛋白的黏附能力,增強基質(zhì)金屬蛋白酶的表達水平,增加腫瘤細胞的侵襲力。進一步研究COX-2選擇性抑制劑塞來昔布與舌鱗狀細胞癌侵襲之間的關(guān)系,對舌鱗狀細胞癌的治療和預(yù)后有非常重要的意義。

[Abstract] Cyclo-oxygenase(COX) is the major rate-limiting enzyme for the synthesis of prostaglandins. COX-2,which is the inducible form, involved in inflammation and cancer incidence. Study found, COX-2 was overexpressed in the majority of malignant carcinomas including tongue squamous carcinoma. In addition, the increasing activity of COX-2 can improve the adhesion capacity of cells and the extracellular matrix protein, enhance expression level of matrix metalloprotease, and increase the invasion of tumor cells. 舌鱗狀細胞癌(tongue squamous cell cacrci-noma,TSCC)是口腔頜面部最常見的惡性腫瘤之一,早期即可發(fā)生頸部淋巴結(jié)轉(zhuǎn)移。舌鱗狀細胞癌轉(zhuǎn)移嚴重影響患者的預(yù)后,也是舌鱗狀細胞癌患者死亡的重要原因之一。大量的研究資料顯示,環(huán)氧化酶(cyclo-oxygenase,COX)-2通過多種機制參與惡性腫瘤的發(fā)生發(fā)展和浸潤轉(zhuǎn)移,而其選擇性COX-2抑制劑塞來昔布對多種惡性腫瘤的發(fā)生和發(fā)展都有明顯的抑制作用。COX-2在舌鱗狀細胞癌中呈過度表達,其水平明顯高于癌前期病損黏膜和正常上皮。下面就該酶在舌鱗狀細胞癌侵襲中的作用及其特異性抑制劑塞來昔布對舌鱗狀細胞癌侵襲作用的抑制作一綜述。
